We have established a unique MS TWIN STUDY and are longitudinally following >100 pairs of monozygotic twins with discordance for Multiple Sclerosis. The aim of this study is to identify risk and trigger factors that differ between cases and controls matched not only for age, gender and genetic background, but also for a broad range of (early) environmental factors. In this high risk cohort we additionally explore early alterations in the immune system during prodromal MS leading to clinical MS manifestation. We use a wide range of methods from clinical phenotyping including innovative MRI imaging, optic coherence tomography and neuropsychological testing as well as a broad spectum of innovative research technologies such as single cell sequencing in CSF, deep microbial profiling and microbiome transfer experiments in a germ-free spontaneous EAE model as well as epigenetics (including scATAC) and genetics.
